Description
A 35 amino-acids peptide with 3 disulfide bridges , synthetic, initially isolated from the venom of the Chinese gold earth tiger spider Ornithoctonus huwena, also known as Cyriopagopus schmidti, Haplopelma schmidti, Haplopelma huwenum.
Biological activity

Huwentoxin IV(HwTx-IV) acts selectively on Tetrodotoxin-sensitive (TTX-S) voltage-gated sodium channels, with an IC50 of 30 nM in rat DRG neurons. It preferentially inhibits neuronal voltage-gated sodium channel subtype hNav1.7 (SCN9A, IC50 is 26 nM), rNav1.2 (SCN2A, IC50 is 150 nM), and rNav1.3 (SCN3A, IC50 is 338 nM). Huwentoxin IV inhibits the activation of sodium channels by trapping the voltage sensor of domain II of the site 4 in the inward, closed configuration. Shows a weak or no inhibition on rNav1.4/SCN4A (IC50=3.9 µM) and hNav1.5/SCN5A sodium channels (IC50=25->10 µM) (PubMed:18628201, PubMed:25658507). This toxin has also been shown to weakly inhibit Kv11.1/KCNH2/ERG1, Kv1.2/KCNA2 and Kv1.3/KCNA3.
